Neethu Vanajakshi was born on 26 February 1993 in Gadag, Karnataka. Raised in that region, she attended Model High School in Gadag-Betigeri and later moved to Bengaluru for further studies. Her formal education includes a BBA from a college in Bangalore and an animation diploma from KEN School of Art, Bangalore. From childhood she showed a strong interest in art, which shaped her later career choices.

Neethu was assigned male at birth and spent much of her early life navigating her gender identity privately. After relocating to Bengaluru, she connected with others from the LGBTQ community, began medical transition with hormonal treatment and surgeries, and changed her name from Manjunath to Neethu Vanajakshi.
